Twenty-five years later, his elder brother <strong>Francesco I de\u2019 Medici<\/strong>, Grand Duke of Tuscany, also died after developing similar symptoms. For centuries, historical accounts attributed both deaths to malaria, although Francesco\u2019s death continued to fuel theories of poisoning.<\/p>\n<p>Now, a new study conducted by the <strong>Division of Paleopathology<\/strong> <strong>within the Department of Translational Research and New Technologies in Medicine and Surgery at the University of Pisa<\/strong>, in collaboration with researchers from <strong>Yale University<\/strong>, provides compelling new evidence and sheds new light on the evolution of malaria in the past.<\/p>\n<figure id=\"attachment_528445\" aria-describedby=\"caption-attachment-528445\" style=\"width: 849px\" class=\"wp-caption aligncenter\"><img fetchpriority=\"high\" decoding=\"async\" class=\"wp-image-528445 size-full\" src=\"https:\/\/www.unipi.it\/wp-content\/uploads\/giovanni_francesco_medici.jpg\" alt=\"\" width=\"849\" height=\"500\" srcset=\"https:\/\/www.unipi.it\/wp-content\/uploads\/giovanni_francesco_medici.jpg 849w, https:\/\/www.unipi.it\/wp-content\/uploads\/giovanni_francesco_medici-300x177.jpg 300w, https:\/\/www.unipi.it\/wp-content\/uploads\/giovanni_francesco_medici-768x452.jpg 768w\" sizes=\"(max-width: 849px) 100vw, 849px\" \/><figcaption id=\"caption-attachment-528445\" class=\"wp-caption-text\">Left: Cardinal Giovanni de\u2019 Medici; right: Francesco I de\u2019 Medici.<\/figcaption><\/figure>\n<p>Published in<a href=\"https:\/\/www.cell.com\/iscience\/fulltext\/S2589-0042(26)01746-3\"> iScience<\/a>, the study analysed DNA extracted from the skeletal remains of the two brothers in search of genetic traces of <em>Plasmodium<\/em>, the genus of parasitic protozoa responsible for malaria. <strong>The analyses identified, in the remains of Giovanni de\u2019 Medici, the DNA of a previously unknown strain of <em>Plasmodium falciparum<\/em>, the species responsible for the most severe form of human malaria<\/strong>. According to the study, this strain carries two unique genetic mutations, which are thought to have emerged during the parasite\u2019s demographic expansion and its spread across Europe.<\/p>\n<p>In the remains of Francesco de\u2019 Medici, the researchers identified <strong>genetic traces attributable to both <em>P. falciparum <\/em>and a second species, <em>Plasmodium malariae<\/em><\/strong>. The presence of two different species in the same individual is consistent with previous research conducted in Belgium covering the same historical period, which documented multiple infections. However, <strong>Alexander Ochoa<\/strong>, first author of the study and Associate Researcher at the Yale Human Evolutionary Genomics Laboratory, explained that further genetic analyses will be needed to confirm whether the two species were indeed co-circulating in central Italy during the 16th century.<\/p>\n<p>\u201cOur study is an excellent example of how modern analytical techniques can be applied to ancient DNA to better understand the past,\u201d said <strong>Serena Tucci<\/strong>, Assistant Professor of Anthropology in Yale\u2019s Faculty of Arts and Sciences and head of the Yale Human Evolutionary Genomics Laboratory. \u201cBut our work goes beyond shedding light on history: it has also generated new data that may contribute to ongoing and future research on malaria, a disease that continues to cause millions of cases and hundreds of thousands of deaths worldwide every year.\u201d<\/p>\n<figure id=\"attachment_528448\" aria-describedby=\"caption-attachment-528448\" style=\"width: 2240px\" class=\"wp-caption aligncenter\"><img decoding=\"async\" class=\"wp-image-528448 size-full\" src=\"https:\/\/www.unipi.it\/wp-content\/uploads\/Francesco-I.jpg\" alt=\"\" width=\"2240\" height=\"1488\" srcset=\"https:\/\/www.unipi.it\/wp-content\/uploads\/Francesco-I.jpg 2240w, https:\/\/www.unipi.it\/wp-content\/uploads\/Francesco-I-300x199.jpg 300w, https:\/\/www.unipi.it\/wp-content\/uploads\/Francesco-I-1024x680.jpg 1024w, https:\/\/www.unipi.it\/wp-content\/uploads\/Francesco-I-768x510.jpg 768w, https:\/\/www.unipi.it\/wp-content\/uploads\/Francesco-I-1536x1020.jpg 1536w, https:\/\/www.unipi.it\/wp-content\/uploads\/Francesco-I-2048x1360.jpg 2048w\" sizes=\"(max-width: 2240px) 100vw, 2240px\" \/><figcaption id=\"caption-attachment-528448\" class=\"wp-caption-text\">The remains of Francesco I de\u2019 Medici.<\/figcaption><\/figure>\n<p>\u201cThe study of ancient DNA not only allows us to diagnose malaria in the remains of individuals who lived in the past,\u201d said Ochoa. \u201cIt also provides a unique window onto the evolution of malaria species, in this case <em>P. falciparum<\/em>, helping scientists better understand how the pathogen adapts over time.\u201d<\/p>\n<p>To carry out the research, the team extracted DNA from four ribs: three belonging to Grand Duke Francesco and one to Cardinal Giovanni. The two brothers are buried in the Medici Chapels, within the Basilica of San Lorenzo in Florence, which houses the tombs of the principal members of the Medici family.<\/p>\n<p>The findings provide new insights into the genetic diversity of <em>P. falciparum<\/em> and contribute to reconstructing the spread and evolution of malaria in central Italy during the Renaissance and the centuries that followed. Malaria was endemic in central Italy from antiquity until the twentieth century, when eradication campaigns succeeded in eliminating the disease from the region. According to the World Health Organization, around 282 million cases of malaria and more than 610,000 deaths were recorded worldwide in 2024.<\/p>\n<p>The new evidence also makes it possible to reinterpret well-known episodes in Medici history in light of genetics. Giovanni de\u2019 Medici contracted malaria together with his mother, Eleonora di Toledo, and his younger brother Garzia during a journey along the Tuscan coast, where marshland areas were known breeding grounds for mosquitoes carrying the disease.
